Ecology of fastidious phytopathogenic bacteria

— 聖誕紅與聖誕紅分枝誘導性植物菌質體之交互作用

聖誕紅 (Euphorbia pulcherrima) 是世界上廣為栽培的盆花。幾乎市面上所有販售的聖誕紅皆具有良好的分枝性,而聖誕紅枝所以能產生這種性狀,主要是因為它們受到了聖誕紅分枝誘導性植物菌質體 (PoiBI) 的感染。除了分枝程度之外,PoiBI 也能改變聖誕紅植株葉片與苞片的顏色與形狀。也因此聖誕紅與 PoiBI 可說是植物病害中,極少數被廣為應用在園藝產業上的特殊案例。

一般聖誕紅育種者大多會先以雜交或是誘變的方式獲得遺傳背景不同的植株,並待植株長成後再植入 PoiBI,才能開始評估植株的性狀並進行品種檢定,其過程相當耗時。本團隊透過分子定量技術搭配扦插繁殖試驗發現,PoiBI 在不同植株間以及不同植株部位中的分佈情形相當不均勻。此外,本團隊也發現 PoiBI 在採穗母株 (stock plant) 中的族群量也與扦插繁殖苗呈現高分枝性的植株比例具正相關性 (Lee et al., 2021)。這些結果顯示,若能有效掌握 PoiBI 與寄主之交互作用,便能更有效率地改良聖誕紅性狀。

此外,本團隊近期也發現 PoiBI 在植株內的族群量與聖誕紅防禦相關基因之表現量呈現顯著的副相關性 (Lee et al., 2022)。未來本團隊將根據這些趨勢進行試驗設計,並針對 PoiBI 與聖誕紅之分子交互作用進行深入探討。

Candidatus Liberibacter 屬細菌與媒介昆蟲之交互作用

Candidatus Liberibacter 能夠在一些重要作物上引起嚴重的病害 (如柑橘黃龍病;citrus greening)。這些病原細菌主要藉由歸屬於木蝨總科的昆蟲進行傳播。雖然近年來國際上有關 Ca. Liberibacter 的研究非常的多,但絕大多數的研究皆著重在少數的作物與媒介昆蟲種類。這樣的情形可能縮限了各界對 Ca. Liberibacter 與木蝨交互作用的認知。本團隊近期在棲息於臺灣木本植物中的木蝨發現了 Ca. Liberibacter 的存在 (Fang et al., 2023; Lin et al., 2022)。目前也發現有些菌種的在媒介昆蟲中的族群量可能會受到昆蟲性別、體色等因子的影響,而有些 Ca. Liberibacter 菌種的感染則可能與木蝨適性有關 。目前本團隊正在針對這個新的系統進行進一步的研究。

Genetic and phenotypic diversity of plant pathogenic bacteria

— Phylogenetic and phenotypic analyses on soft rot bacteria in Taiwan

屬於 Pectobacteriaceae 的各種軟腐細菌為相當常見的植物病原。過去臺灣分離到的軟腐細菌主要被區分為 Erwinia carotovora subsp. carotovora (Ecc) 與 Erwinia chrysanthemi (Ech)。隨著分子遺傳學的發展,目前國外已針對這些細菌的分類系統進行大幅的修改,而過去歸屬於 EccEch 的菌株目前已分別被歸類至 PectobacteriumDickeya 的不同物種當中。過去尚未有研究針對臺灣的菌株依照新的分類系統進行詳細的分析,也在病害診斷與植物檢疫方面衍生出了許多困擾。為了對臺灣軟腐細菌的分類地位與特性能有更進一步的認識,本團隊近年開始針對過去由臺灣田間分離到的菌株進行了系統發生學與表型分析。

本團隊近期已針對臺灣的 Dickeya 屬進行了相關的分析。試驗結果顯示臺灣至少可分離到 5 個不同的物種,而部分菌種似乎具有一定程度的寄主偏好性。強勢接種試驗發現,這些細菌的致腐能力在物種間與物種內皆具有高度變異性,而這些細菌在不同植物寄主中的致腐能力也有所不同 (Wei et al., 2021)。目前本團隊也開始針對臺灣分離之 Pectobacterium 菌株進行相同的分析 (Tang et al., 2024; Wang et al., in preparation),期望這些試驗成果能幫助提升各界對臺灣軟腐細菌的生態與多樣性之了解,進而增進病害診斷與防治的精準度。

Discovery of novel bacterial plant pathogens

本研究室持續在田間搜尋過去未知之植物細菌。目前已成功發現與發表屬於 Xanthomonoas euvesicatoria 之全新病原型,全名為 Xanthomonoas euvesicatoria pv. polysciadis (Wang et al., 2023)。

Diagnosis of emerging bacterial diseases in Taiwan

本研究室致力於研究與鑑定臺灣新發現之植物細菌性病害。相關研究內容包括以生理生化測試搭配系統發生分析鑑定新發現之病原,或是進一步研發病原細菌之分子檢測方法,或是篩選有效的防治藥劑。本團隊近年已陸續在多種花卉與蔬菜作物報導多種由 XanthomonasDickeyaPectobacteriumCandidatus Phytoplasma、Robbsia 等細菌所引起的病害 (see publication list below)。
