目前研究方向

(1) 植物病毒分子生物學

本實驗室在 PRSV 之研究居世界先導地位,除早期分類及血清學之研究外,於 1992 年即發表 PRSV RNA 全長 10326 個核甘酸之序列,並解析各基因之組成。據此,我們已先後完成其鞘蛋白基因轉殖木瓜,試管內及生體內具感染力轉錄體之構築。目前正進行病毒在寄主專一性、蛋白裂解過程、病原性強弱、以及交互保護及轉基因木瓜專一抗病性等相關基因分析。具感染力的病ZYMV載體毒還可進一步用來表現外源蛋白,本實驗室利用這種技術已可成功純化外源蛋白並用於血清製備或是過敏反應的過敏純化,該研究並投稿於國外免疫學領域頂尖期刊中。此外,本實驗室在 Tospovirus 屬之地位佔有重要國際地位,除了先後發表兩種新病毒種 WSMV 及 PCFV 之外,並且亦完成此二病毒基因體核酸序列分析及類緣定位,而本實驗室主持人 葉錫東 教授為國際 tospovirus 屬七位分類委員之一。

與洛克斐勒大學合作研發利用人工改造微小 RNA (artificial microRNA)的技術,帶有該人工改造微小RNA轉殖的植株可成功對抗蕪菁嵌紋病毒(TuMV)及蕪菁黃化嵌紋病毒,此種技術之優點是基因構築時間短,所帶有的病毒序列也只有21鹼基對,抗病效果於低溫下仍具有效用,目前正研發利用其來發展廣泛性轉殖作之應用。

(3) ) 病毒鞘蛋白轉基因植物 (內含成果照片展示,按一下標題可進入觀賞)

1986 年 Powell Abel 等人首先將菸草嵌紋病毒 (TMV) 的鞘蛋白基因轉殖到菸草細胞內,經再生過程得到表現此基因的菸草,其子代經挑戰接種 TMV,發現有延遲發病之效果,此現象命名為鞘蛋白媒介的抗性 (coat protein-mediated resistance)。近年來,鞘蛋白轉型植物在許多病毒群中皆有報告發表。本實驗室已完成木瓜輪點病毒 (PRSV) 鞘蛋白轉基因木瓜,且經挑戰接種 PRSV,證實此轉基因木瓜具有抗病特性,可以有效的防治 PRSV。而本實驗室是國內第一個取得轉基因作物田間試驗許可的研究室,目前正進行商品化評估。此外,其它轉因植物包括 PRSV W 型鞘蛋白轉基因甜瓜與西瓜、ZYMV 鞘蛋白轉基因甜瓜及 WSMV NP 蛋白轉機基因甜瓜等亦正構築中。

(前利用抗血清診斷病毒病害為一種經濟快速的方法,因此本實驗室亦自行生產各種病毒之多元與單元抗體,以利病害診斷與各項研究工作之進行。另外上述 Potyvirus 及 Tospovirus 屬病毒的核酸探針以及 RT-PCR 檢測系統亦均有立4) 輕症病毒交護保護系統

交護保護作用之現象於 1929 年首先由 Mckinney 發現,即植物受一病毒感染後,對同類型病毒具有免疫力。利用此現象,可先讓植物感染一種對其本身無不良影響或影響不大的輕症病毒,此植物受輕症病毒的保護對同類型的強系病毒則有抗病能力,而達到防治的目的。本實驗室利用亞硝酸誘變獲得 PRSV 輕症病毒,並用於防治 PRSV 成效卓著,蜚聲國際。本實驗室並已完成 PRSV 夏威夷品系、PRSV 台灣品係及 PRSV 夏威夷弱系病毒生體外及生體內具感染能力轉錄體。此轉錄體可以提供研究 PRSV 病毒核酸各基因之功能,並且可以藉由研究這些病毒轉錄體產生各種弱系病毒以提供交護保護作用。

(5) 病毒診斷工具之製備
